Clinical & biological endophenotypes were tested as potential mediators of the effect of genotype on suicide risk. 486 subjects with mood disorders & 119 healthy volunteeers, all European Caucasians, were genotyped for COMT-V158M & assessed for psychiatric diagnoses, lifetime suicidal behaviour, & lifetime impulsivity, hostility, & aggression. CSF monoamine metabolites were assayed in a subsample of 111 mood disorder patients. No association was found between genotype & mood disorder diagnosis or with reported history of suicide attempt in mood disorder subjects. There was no association between genotype & lethality or method of suicide attempt, or with aggressive/impulsive traits. There was no difference in monoamine metabolites by genotype. (46 refs.) JA
